    Perhaps the most exciting area of current research involves the potential neuroprotective effects of olive oil polyphenols. Scientists are investigating whether these compounds might support brain health and cognitive function, particularly as we age.

    Crossing the blood-brain barrier

    For any compound to affect the brain directly, it must cross the blood-brain barrier - the selective membrane that protects the brain from potentially harmful substances in the bloodstream. Research has confirmed that olive oil polyphenols, including oleocanthal and hydroxytyrosol, can cross this barrier, allowing them to exert effects directly within brain tissue.

    The Beta-Amyloid research

    One line of investigation focuses on beta-amyloid, a protein that accumulates in the brains of individuals with Alzheimer's disease. Research published in ACS Chemical Neuroscience found that oleocanthal enhanced the brain's natural clearance of beta-amyloid by upregulating transport proteins that help remove these proteins from the brain. This mechanism is particularly intriguing because it suggests oleocanthal may support the brain's own housekeeping systems rather than simply acting as a passive antioxidant (Abuznait AH, et al. ACS Chem Neurosci. 2013;4(6):973-982).

    The Temple University study

    Researchers at Temple University published findings from a carefully controlled animal study in 2017. Mice fed an extra virgin olive oil-enriched diet (equivalent to regular human consumption) showed improved performance on memory and learning tests, preserved synaptic integrity (the connections between brain cells), increased autophagy activation (the cellular "cleanup" process that removes damaged proteins and organelles), and reduced beta-amyloid pathology. The researchers specifically attributed these effects to the polyphenolic compounds in the olive oil, not merely its fat content, as control oils did not produce similar benefits (Lauretti E, et al. Ann Clin Transl Neurol. 2017;4(8):564-574).

    Systematic reviews and meta-analyses

    As evidence accumulates, researchers conduct systematic reviews to evaluate the totality of findings. A 2024 systematic review published in the International Journal of Molecular Sciences examined all available studies on extra virgin olive oil in Alzheimer's disease, comprehensively reviewing the neuroprotective mechanisms of olive oil polyphenols including their antioxidant, anti-inflammatory and anti-amyloid properties (Alkhalifa AE, et al. Int J Mol Sci. 2024;25(3):1914).

    A 2025 systematic review in Current Neuropharmacology evaluated the effect of oleocanthal - a key polyphenol in extra virgin olive oil - on amyloid-β burden in preclinical models of Alzheimer's disease, finding that oleocanthal-supplemented diets consistently supported the clearance of amyloid-β across the blood-brain barrier (Zupo R, et al. Curr Neuropharmacol. 2025;23(10):1249–1259).

    A 2025 review in Frontiers in Nutrition examined the neuroprotective properties of extra virgin olive oil polyphenols more broadly, discussing mechanisms including antioxidant effects, anti-inflammatory activity, mitochondrial support and benefits for brain metabolism (Wei L, et al. Front Nutr. 2025;12:1736633).

    Research on other polyphenolic compounds has demonstrated effects on brain and behaviour, supporting the broader concept that dietary polyphenols may influence cognitive function (Poulose SM, et al. Neurochem Int. 2015;89:227-233).

    Human Evidence: The PREDIMED-NAVARRA Trial

    While much brain health research involves laboratory and animal studies, human evidence is emerging. The PREDIMED-NAVARRA substudy assessed cognitive function in 447 participants at high cardiovascular risk. Those assigned to the Mediterranean diet supplemented with extra virgin olive oil showed improved cognitive performance on neuropsychological tests compared to the control group, with benefits most pronounced in memory and frontal cognition domains (Martínez-Lápiscina EH, et al. J Neurol Neurosurg Psychiatry. 2013;84(12):1318-1325).

    What this means

    While more human clinical trials are needed, the converging evidence from laboratory studies, animal research and initial human trials suggests that olive oil polyphenols may support brain health through multiple mechanisms. This represents one of the most promising areas of ongoing nutritional neuroscience research.

    Oleaphen's concentration of 2,236 mg/kg provides exceptionally high levels of the specific polyphenols - oleocanthal, hydroxytyrosol, and oleacein - that researchers have identified as potentially beneficial for cognitive health.
